Dynamic Presentation Course

Assalamu'alaikum

Now just finished from attending a course which I took before another course on next week and what was it about??? yah... I think you can guess it from the title I gave... lol but now I will tell it everything about it in my 'mother' language.... cos I think it will be better and not much effort to write on it.... (just so lazy after had to use English everytime... and had to find proper words during the course... tough then I think I still didn't speak English well but nevermind... as long as I try... and try... it will improve by itself, won't it? Ok... now start to switch the language... one... two... three... here we go


Akhirnya setelah dua hari ikut kursus tentang presentasi, sebenarnya lebih tepatnya training karena ya semacam training.... ada practical thing-nya lah... jadi bukan hanya duduk ndengerin teori-teori... (seperti satu pepatah... teori aja sih bisa... )... nah trainingnya ini dimulai sejak kemaren (05 Juni, red) dari jam 9 waktu Groningen tepat (alias 9 a.m. sharp) tapi tetep yang namanya telat ko masih ada juga.... heran... heran... tapi yang pasti sih bukan karena pesertanya jadinya mulai telat... so no one is blamed... and just accepted it... there will always be like this... wherever you are though sometimes do not really like at that point. loh...loh... ko jadi ke bahasa Inggris lagi sih hehehehehe... ya udah deh.. campur-campur aja ampe mana ya tadi... tunggu bentar.. baca lagi.... hmhmhmhmhm... oh ya tentang telat... walopun telat dikit sekitar 10 menit-an... tetep dibilang telat.. ya ga... nah mulai lah kita duduk di masing-masing tempat (kalo ini ga diatur... pilih sesukanya tapi kayanya udah ga jaman kali ya... duduk aja harus diatur hehehehehe).... setelah itu kalo ga salah mulai saling kenalan (satu orang aja... karena agak mendetail seperti educationhistory-nya... etc...) nah abis itu mulai lah kita memperkenalkan orang yang baru kita kenal itu di depan kelas... yang paling kagetnya (dramatisir nih ceritanya hiihihihihihi... ) setiap orang yang memperkenalkan itu direkam di video... nantinya dibahas.... walah...walah... kebayang kan gimana ngliat diri sendiri... heheheh (walo ga kaget lagi sih ama yang namanya kamera... la selebritis gitu loh hahahahahahhaahah pasti kamera ada dimana-mana).

Oya ampir lupa siapa aja pesertanya... dari 10 peserta ada dua orang dari TOP master yang ikut kursus ini... siapa aja kah mereka... yang pasti sih yang nulis blog ini sendiri... si "lucu" tea terus ada temen satu lab, namanya Jolanda van Muenster n yang pasti udah tau kan orang mana dari namanya... terus ada Ite yang ngambil program PhD di Zernike... lupa di lab yang mana tapi penelitiannya tentang protein P450ssc, trus ada Jan Peter yang orang Jerman tapi sekarang ngambil PhD di Groningen dengan kerjaannya yang berbau nanovalve dan masih berbau bio. Selanjutnya ada Magdalena, orang Polandia yang kerjanya di satu gedung di Haren tapi grupnya grup Molecular Microbiology, trus ada Araz orang Azerbaijan yang lama di Turki dan masih di satu gedung tapi lupa dari grup mana. Last but no least, dua orang dari TOP Master, Barbara Sanders dan Bangwen. Setelah perkenalan selesai baru mulai sedikit teori.... sedikit exercises... alias latihan pengolahan suara terus tubuh... karena suara yang keluar juga dipengaruhi dari gerak tubuh. Selanjutnya barulah dimulai ama presentasi pertama masing-masing peserta dengan topik yang jauh dari kerjaannya sekarang... ya kalo nge-lab ya jauh dari topik labnya n semacam itulah. Kebanyakan presentasi kali ini tentang tempat yang akan mereka kunjungi atau yang telah dikunjungi selama liburan, seperti ada yang cerita tentang singing giant waktu dia liburan di Maui, Hawaii baru-baru ini (wah... Hawaii.. mauuuuuu.... tapi kapan ya.... and sama siapa ya... hehehehehehe ), ada satu juga yang berbeda... dia presentasi-in tentang binatang kesukaannya yang apalagi kalo bukan kucing... kucingnya bener-bener lucu ampe dijadiin sebagai objek di slide-nya... jadi pengen punya... sayangnya ko ga mungkin ya melihara di sini... wish have own house  terus ada yang sama-sama merawatnya (duh... duh... ko jadinya ngelantur gini yah.... hehehehehe... semoga belum masuk tahap kronis )... oya kalo diriku sendiri mresentasiin tentang pisang aroma... dari bahan-bahan untuk membuatnya ampe ke cara membuatnya... abis udah ga kebayang mo presentasi apa... ya udah di pagi harinya bikin slide tentang pisang aroma... yang bener-bener kurang persiapan... masa slide cuma 3 biji walo emang cuma 3 menit sih waktunya... tetep aja... kalo udah presentasi yang ada tancap gas mulu... jadi ngomongnya cepet... dan waktunya pun ga nyampe 3 menit, sekali lagi semua presentasi ini direkam dan dibahas satu-satu setelah semua selese... yang pasti sih... untuk diriku... dari gurunya bilang presentasinya sangat antusias walo harus diatur vowel-nya (alias struktur pengucapan kata ) katanya sih kadang-kadang suka ada suku kata yang tiba-tiba menghilang atau kurang strecth... yah ga papalah... namanya juga masih dalam tahap belajar untuk jadi lebih baik... (cie... cie... ) setelah itu baru ada PR and presentasi buat besok (06 Juni, red) tentang kerjaan masing-masing...

Keesokan harinya (alias hari ini, Jumat 06 Juni) kerasanya ko males ya pergi kursus... apalagi nanti harus presentasi lagi (ya iyalah namanya juga dynamic presentation course ) ga tau aja ko rasanya bakalan terjadi sesuatu hal... tapi tetep harus semangat demi perbaikan kemampuan presentasi dan seperti hari sebelumnya, kursus dimulai jam 9... kali ini bener-bener jam 9 tepat.... dan dimulai dengan bincang-bincang mengenai kesulitan selama presentasi... yah pokonya segala kesulitan yang berkaian ama presentasi entah dari slide-nya trus persiapan diri ampe saat presentasi-nya sendiri. Setelah itu mulai ada exercise lagi, cuma kali ini kita disuruh mengambil satu lirik atau bait boleh dari lagu atau puisi lalu diekspresikan dengan berbagai macam ekspresi, dari marah, malu, jadi orang mabuk, marah ampe ke ekspresinya orang gila (wah... bener-bener rame deh...). Setelah latihan ekspresi, barulah kita dikasih satu task untuk memperagakan suatu kejadian tapi dengan kata-kata dari satu bait yang kita ambil sebelumnya.

Nah kalo diriku kebagian task untuk memperagakan saat menerima penghargaan Oscar (mimpi yang udah lama akhirnya terjadi... walo cuma pura-pura ) terus ngucapin terima kasih ke semua pihak yang sudah membantu jadi sukses sperti itu dan berterima kasih ke temen yang meninggal 3 minggu sebelum acara ini berlangsung dan penghargaan Oscar itu ditujukan untuknya... yang paling sulit sih saat ngomongnya karena harus pake kata-kata dari lirik yang kita ambil sebelumnya... dan... lirik yang aku ambil adalah.... lirik dari lagu Lelaki Buaya Darat... jadi bingung sendiri juga abis ga pantas... yah yang penting sih ekspresi... seperti iklan salah satu produk (apa ya??? lupa)... ekspresinya mana... ekspresinya... dan seperti sebelumnya... tugas ini juga direkam... feel like superstar  setelah selesai semua dan sedikit bincang-bincang lagi... mulailah tugas utama yaitu presentasi dari bidangnya...... huaa.......... ko tetep aja pabaliut ya ngomongnya (eh ga juga ding...) cuma ada yang kelupaan yang harus diomongin plus kurang general ngejelasinnya... jadinya ga ps buat audience-nya hiks... hiks... tetep aja kalo udah nervous apapun yang udah dipersiapin di otak ga kesampean ke mulut untuk diucapkan... sedihnya... ga sinkron antara otak dan mulut. But sekali lagi... ini kan latihan buat jadi lebih baik lagi... cuma ada satu kejadian lagi yang bener-bener bikin malu setengah mati dan kerasa bodo banget... waktu disuruh jelasin selama 2 menit tentang suatu tema yang ditulis sama kita masing-masing setelah itu diacak... ada satu bagian yang hanya liat judul... langsung ngomong di depan... walah... udah ada persiapan aja masih suka pusing ama kata-kata apalagi tanpa persiapan... hua.... bener-bener jadi malu and kerasa ko bodo banget... (ooopss.. maaf ya 4JJ I... ga bermaksud menghujat diri sendiri tapi... ya... gitu deh... seseorang perfeksionis yang jarang untuk memberikan dirinya penghargaan)..

After all... the most important thing is... now realize more... which side I'm good enough and which side I have to improve and work more on it... So thank you to all of u... Jolande and Wytze from Ludens company... who gave us... more knowledge how to do good presentation... and also advices that u gave to all of us during course.

Pores in Granular Starches versi Indonesia

Kayanya perlu juga nih sekali-sekali nulis dalam bahasa "Ibu" biar mudah dimengerti hehehhehe , nah sekarang yang mo dibahas ya apalagi... kalo bukan hal penelitian yang sebelumnya udah di-post seminggu yang lalu cuma sekarang bedanya cuma di-bahasa plus mungkin agak lebih lengkap di sini, mudah-mudahan lebih mudah dimengerti .

Penelitian yang sedang dilakuin ampe saat ini topiknya adalah enzim, alpha-amylase, dari mikroorganisme Microbacterium aurum yang mampu mendegradasi -mengolah- pati (pati mentah atau raw starch -granular starch-). Kenapa pati mentah dan kenapa mikroorganisme ini yang digunakan? Satu-satu akan dijelaskan walopun mungkin ga akan terlalu mendalam.

Kenapa pati mentah? karena banyak industri yang menggunakan pati sebagai bahan dasar dalam pembuatan berbagai macam produk terutama produk makanan. Terus apa tuh pati? Pati itu merupakan salah satu sumber karbohidrat yang melimpah dan biasanya tersimpan dalam salah satu organel -bagian dari sel- tumbuhan, amiloplast, dengan bentuk dan ukuran yang bergantung pada jenis tanamannya. Amiloplast ini tidak mudah larut dalam air serta tidak berpori. Dipilihnya kentang sebagai topik utama dikarenakan Belanda merupakan salah satu produsen dan pengonsumsi kentang (soalnya penelitiannya kan di Belanda ), selain itu pula kentang memiliki struktur yang kaku dan bersifat kristalin. Enzim yang ada hingga saat (yang banyak digunakan) adalah enzim yang mampu mendegradasi pati yang telah terlarut, pati yang telah dimodifikasi dan biasanya dimodifikasi dengan menggunakan pemanasan sehingga berbentuk seperti gelatin. Enzim kebanyakannya bekerja maksimum dalam suhu yang relatif menengah (meso). Proses gelatinisasi ini tentunya menggunakan energi yang sangat besar (suhu tinggi) sehingga kemudian digunakan enzim yang termostabil, enzim yang tahan dan dapat bekerja di suhu tinggi, oleh karena itu enzim-enzim seperti ini yang kemudian banyak diupayakan oleh para saintis.

Namun walau demikian, tetap saja dibutuhkan energi yang cukup besar untuk pengolahan pati. Dengan adanya proyek ini, enzim yang mampu mendegradasi pati mentah, diharapkan mampu mengurangi bahkan meniadakan penggunaan suhu tinggi dalam proses pengolahan pati, sehingga energi yang dibutuhkan lebih sedikit dan menghemat biaya.


Nah... alhamdulillah, minggu lalu (sebenernya sih udh dapat sejak akhir taun lalu, tapi dicoba reproducible ato ga)... terbukti dengan jelas lewat SEM... kalo supernatan dari mikroorganisme yang diinkubasi dengan beberapa macam pati mampu mendegradasi pati-pati tersebut, salah satunya dengan terbentuknya pori-pori pada partikel-partikel pati (Gambar atas kiri, kentang yang regular; gambar atas kanan, kentang yang diinkubasi dengan supernatan M. aurum B8.A; gambar kiri, kentang yang diinkubasi dengan supernatan M. aurum DSMZ 8600, mikroorganisme dari supplier bakteri di Jerman). Selain itu, aktivitas yang ada merupakan aktivitas enzim yang unik karena juga udh dikonfirmasi pake mikroorganisme yang sama genus-nya dan ternyata tidak dihasilkan aktivitas yang sama. So... hip...hip... huurayyy... alhamdulillah... bisa juga dapat hasil yang diinginkan walo harus menunggu kurang lebih 10 bulan fiuhh.... tapi belum bisa bernafas dengan lega... masih banyak kerjaan yang harus dituntaskan.... wish me luck sekarang sedang berusaha untuk memodifikasi enzim itu sehingga tau apa yang menyebabkan enzim itu bisa membuat pori-pori.

Pores in Granular Starch

Hi...hi... how's going? well... here... at least for these weeks, weather seemed so nice... most of days were sunny even though it's quite cold. But never mind, cos now I will tell another thing which is about my research here, in Groningen. I think I've never tell anything about I will do and have done on my research since I was in April 2007. So... let the story begins...

My research project topic is structure/function relationship of alpha-amylase type of enzymes: new reaction specificities, and functional roles of carbohydrates binding modules. Hohohoho... sounds interesting, doesn't it? well... in short way, it's about enzymes of microorganism's activity on granular starch. And why it's so interesting topic? I will explain it one by one... so it can be understandable as well for anyone who's not in the same subject

My work for more less 10 months was on enzymes of Microbacterium aurum which can degrade granular starch, on potato granules starch as main target. and why do we focus on starch especially on potato granules starch? Short explanation, cos I think most of you know what is starch, starch is glucose polymer which is a long term storage compound present in seed, roots, and tubers of many plants. Starch is stored in amyloplasts (figure above, amyloplast of potato, credit to Wikipedia), which are water insoluble granules that vary in shape  and structure depending on the botanical origin. Potato starch granules are rigid, have a high crystallite and do not possess pores. Therefore, potato starch is rather resistant to microbial degradation at low temperatures. Moreover, there have a few reports about this genus (Microbacterium) on enzymes specifically.

Why are we looking for activity on granular starch at low temperature? As we know, there are a lot of food industries which use starch as a source to get many products from it. And before it was processed into several intermediate products and final products, it was gelatinized by swelling in high temperature. If this step could be reduced from this process, it will reduce of use of energy which is more economize .

So... from my research, I found that culture fluids of M. aurum B8.A can digested granular starch from wheat, tapioca, and potato starch. Although a high level of activity was found with wheat and tapioca granules starch, digestion of potato granules starch was slow. Scanning Electron Micrography (SEM) figures showed the effect of enzyme(s) on granular starch which gave high density pores on granular. Activity on granular starch was compared to other strain of M. aurum (M. aurum DSMZ 8600) as well, and showed that activity on granular starch was only found in M. aurum B8.A.

Another month... another try hard

Assalamu'alaikum

It is already in 8th day of August, but it seems I can't still get my good performance in my experiment (research)... everything is still going unexpected... yah...actually not everything cos some other things work nicely, but then... one thing, the most important thing... I can't still get it.

So.... now I just want write down what I've done in my experiment, even my protocols... so If there is anyone... know about this protocol, and could give me some advice... it is very welcome... and I'll be really gratefull.

One thing which I really wanted most is to get genomic DNA of Microbacterium aurum (this is microbe which I use in my research). First time... in maybe April or May... I try to isolate that DNA, and I didn't get, but I tried it more... and finally I got one... so I could move into another step of research... then I found that my DNA (I mean that genomic DNA) is not enough or not suitable for next step... so I have to isolate it more. Then... until today (after  maybe 7 or 8 times try to isolate)... I can't get any. First, I think it should be problem in incubation time, then I try a little bit longer (from 15 min to 1 hour)... and still nothing happen (no precipitation which indicate there is DNA in solution)... Second, Lysozyme, enzyme for lysis cell... I try to make it fresh... still nothing happen... I also try to add it more in solution, so... could anyone help me??? (yah... if someone read it... certainly)

Here is my protocol:
1. 100 mL of overnight culture, centrifugated 10 min, 4000 rpm
2. Resuspend cell with 7.5 ml of T10E1 buffer containing 10 mg of lysozyme and incubate for 1 hour at 37 Celcius degree
3. Add 1.5 mL of 10% SDS and 3 mL of 5M NaCl, homogenize
4. Add 0.3 ml of RNAse (10 mg/mL) and incubate for 30 min 55 degree
5. Add 75 mikroliters of Proteinase K (10 mg/mL) and incubate for 30 min, 55 degree
6. Add 2.7 mL of Natrium perchlorate and 18 mL of 24:1 chloroform/isoamyl alcohol, incubate in 4 degree (ice bath) with gently shaking
7. Centrifugation at 10.000 rpm for 10 min, 4 degree
8. Pipet upper phase (15 mL) and precipitate with 2 vol of 100% EtOH
9. Wash three times with 70% EtOH and evaporate EtOH which left in DNA
10. Add sterile water for further experiment

So... this is my protocol, which I've done and success for only once.... wish.. it wil work in my next try... aaamiiiinnnn

Presentation... Done... Not Good Enough

My presentation was finally... done (on Monday 07 May 2007), but... with unexpected... impression from all of the audiences. I mean, actually I think I wasn't doing well....... maybe could said that I was also disappointed for what I've done. My presentation was talked about my research proposal (Raw Starch-Degrading Enzymes of Microbacterium aurum strain), I think I have prepared all the things well... ya... much prepared... for my presentation file (slides)... but not for myself... for speak in front of new colleagues. I always get nervous... if I have to speak in front of many people :( don't know why... never feel comfortable... even though I know what I have to say.
Most people said that was ok... cos it's my first time.... bla..bla...bla... of course that was my first time... but surely... (thanks for make me feel better... I don't mean not to respect for what they've said... also don't wanna complain for all of those things...) I didn't hope like that cos I think I'm ready for it.
But... if I'm not doing something good enough... I don't know... I feel so bad... and think those things much... (maybe cos of perfectionist type)... so... those could make me down... for one moment (hope not take long time ^_^)... these are happen not only once but several times... so... is there something wrong with me?
I think... I have to learn from all of my mistakes...and do better next time...
So... WISH ME COULD BE BETTER ONE ^_^

Nervous...get nervous

Asssalamu'alaikum

Wuaaaa..... it's just two days left... before my first speech in front of my group... Still preparing all of things... but why... can't concentrate just for it :(
It's all about my research proposal presentation... just get nervous on it... I have my presentation file... but it's not complete yet... still add or delete... things.... wuaaaa....dunno it will be on time... cos I have to send it first to my supervisor to be checked.... I think... I have to be on time... right!?
Wish me luck...wish me luck... and can pass it smoothly :)

Biotechnology Seminary

For 2 days I haven't been in my place, though I have a lot to do in lab. But on those days, I think I got a lot of things, new knowledges, friends, etc. Those two days (15-16 November) I've been in Cibinong, LIPI, attended seminar with theme "Capturing Opportunities Through Biotechnology". Actually, we planned just to attend this seminar only on first day... but then... we changed it... and... attended all day ^_^

Oopss.... who are we?? Hmhmhm.... should I describe.. who are we?? Hmhmhm... I think... I won't hehehehe ;p ...... In there, there are Prof Jackson from University of Melbourne; Dr Jefferson from BiOS (Biological Open Source), told about Open Source and patent; and then Dr. Shimizu from Japan, he presented his research on Poliovirus... why Indonesia got infected after free-polio case 10 years.

Und dann... moechte ich andere Erzaehl erzaehlen hehehe ^_^ moechte ich??? Hmhmhm... Ich denke nach... ich moechte nicht....

GS Lecture 2006

Assalamu'alaikum ^_^

Today... just like other days before... we have to work harder than before. This event called The Gruber-Soedigdo Lecture 2006. It will be held on 20-22 June 2006 and will have 2 session, which are seminar on 20 June and workshop on 21-22 June. This event is collaboration between ITB and RuG (Rijkuniversiteit Groningen).

First time, our Minister of Research and Technology will be attend this event... but in this H-1, he considered to cancel attend and give order another to replace him. :(

Seminar entitled is Alternative Fuel: from Renewable Resources to Biofuel and Workshop is Carbohydrate Acting Enzyme: Structure, Modelling, and Application. I wish, this event will success ... Aaaamiiinn... ^_^
